چکیده

Abstract Sex determination is a key developmental event in all organisms. The pathway that regulates sexual fate has been well characterized at the molecular level model free-living nematode Caenorhabditis elegans . This study aims to gain preliminary understanding of sex-determining pathways plant-parasitic Meloidogyne incognita , and extent which roles sex genes are conserved hermaphrodite species, C. M. In this study, we targeted two orthologues, sdc-1 tra-1 from using RNA interference (RNAi). RNAi was performed by soaking second-stage juveniles solution containing dsRNA either Mi-tra-1 or Mi-sdc-1 both. To determine effect target genes, treated with were inoculated onto susceptible cultivar cowpea grown nutrient pouch 28 °C for 5 weeks. development nematodes analyzed different time points during growth period compared untreated controls. Our results showed neither nor have significant role regulating However, silencing significantly delayed maturity adult females but did not affect egg production mature females. contrast, downregulation transcript resulted reduction both single combinatorial RNAi-treated nematodes. indicate may adopted divergent function distinct spp. might an essential female fecundity promising root-knot (RKN) management host-delivered RNAi.

The role of sdc-1 in the sex determination and dosage compensation decisions in Caenorhabditis elegans.

Our previous work demonstrated that mutations in the X-linked gene sdc-1 disrupt both sex determination and dosage compensation in Caenorhabditis elegans XX animals, suggesting that sdc-1 acts at a step that is shared by the sex determination and dosage compensation pathways prior to their divergence. In this report, we extend our understanding of early events in C. elegans sex determination an...

Analysis of the role of tra-1 in germline sex determination in the nematode Caenorhabditis elegans.

In wild-type Caenorhabditis elegans there are two sexes, self-fertilizing hermaphrodites (XX) and males (XO). To investigate the role of tra-1 in controlling sex determination in germline tissue, we have examined germline phenotypes of nine tra-1 loss-of-function (lf) mutations. Previous work has shown that tra-1 is needed for female somatic development as the nongonadal soma of tra-1(lf) XX mu...

Reproduction of Mi-Virulent Meloidogyne incognita Isolates on Lycopersicon spp.

Selection of detectable numbers of Mi-virulent root-knot nematodes has necessitated a greater understanding of nematode responses to new sources of resistance. During the course of this research, we compared the reproduction of four geographically distinct Mi-virulent root-knot nematode isolates on three resistant accessions of Lycopersicon peruvianum. Each accession carried a different resista...

Isolation, cloning, and characterization of a cuticle collagen gene, Mi-col-5, in Meloidogyne incognita

Cuticle collagens form a major part of the nematode cuticle and are responsible for maintaining the overall shape of the animal and its protection from the external environment. Although substantial research on cuticle collagen genes has been carried out in Caenorhabditis elegans, their isolation and characterization in plant parasitic nematodes have been limited to a few genes only. In this st...

Reproduction of Virulent Isolates of Meloidogyne incognita on Susceptible and Mi-resistant Tomato.

The reproductive potential of natural and laboratory-selected Meloidogyne incognita isolates virulent against the tomato Mi resistance gene, all derived from a single egg-mass, were compared when the nematodes were inoculated on susceptible and resistant tomato. Fewer second-stage juveniles (P = 0.01) of the two virulent populations selected under laboratory conditions matured to females on the...
